In the latest study, Xu Shuhua's research team at the Institute of Computational Biology, Chinese Academy of Sciences and Shanghai Academy of Sciences revealed for the first time the difference in transcription levels between Han and Japanese people, and estimated the contribution rate of genetic variation to differences in the gene expression profile of the population. Relevant experts believe that the study will help to understand the relationship between gene expression and genetic variation, and provide clues to elucidate the molecular mechanism of phenotypic differences between closely related populations. Related research results were published online in the Journal of Medical Genetics recently.
The Chinese Han and Japanese have a common genetic origin, and there is also a high degree of similarity in appearance. But the general impression is that the difference in appearance between the Han and the Japanese seems to be recognized by the human eye. So, from a biological point of view, what are the differences in traits between Han people and Japanese? How many differences are caused by environmental differences? How many differences are caused by genetic factors?
Xu Shuhua said that it is not easy to answer these questions. A major difficulty is how to measure and quantify the visually visible final phenotype, which is particularly challenging among close-knit groups. Therefore, the research team decided to start with an intermediate phenotype that can be quantified on a large scale-gene expression profiling, and systematically and carefully analyze the genome-wide expression profiling chip data of the Han and Japanese people.
Through comparative analysis, the researchers identified genes with high expression differences between the two populations, and found that some of the differentially expressed genes are related to human blood and biochemical characteristics, and some genes are related to human height and certain diseases.
In addition, the researchers further estimated the contribution rate of genetic variation to gene expression differences. The results show that for gene expression that has significant differences between Han and Japanese people, the contribution of a single genetic variation is between 7% and 43% . The results of these studies indicate that the differences between the Han and the Japanese are not only differences in geography, language, and culture, but also systemic differences in biology.
